What are the benefits to Top Reading versus Bottom Reading on my fluorescence microplate reader?
Top reading usually provides better signal-to-noise ratios for solution-based assays such as DNA or protein quantification. Bottom reading should provide better results when working with adherent cells, on assays such as GFP gene expression. If in doubt, you can run both Top and Bottom measurements in the same protocol and compare the results to make your final choice.
